"abstract": "Natural bacterial communities (NatComs) represent a promising frontier in sustainable crop protection, yet strain incompatibility and population instability during subculturing remain major bottlenecks in developing reliable bio-inoculants. To address these challenges, we investigated how grapevine scion genotypes with contrasting susceptibility to downy mildew, Chardonnay as susceptible and Voltis resistant, rhizocompartments (endosphere, rhizoplane, and rhizosphere soil), and growth media govern the temporal stability and assembly of culturable NatComs. Using high-throughput SYBR® Green I flow cytometry across successive subcultures, we demonstrated that NatCom stability is largely resilient to environmental shifts, with 87.5% (42/48) of consortia maintaining consistent population densities regardless of host genotype, rhizocompartment, or medium. However, 16S rRNA gene profiling revealed that host genotype exerts a primary selective force on community assembly. Chardonnay NatComs were characterized by co-occurring amplicon sequence variants (ASVs) of Enterobacter and Serratia, whereas Voltis selectively assembled taxa belonging to Achromobacter, Pseudomonas, and Serratia, frequently accompanied by key satellite genera (Rahnella, Stenotrophomonas, Lysinibacillus, and Paenibacillus). Crucially, most NatComs cultured in tryptic soy broth (TSB) significantly enhanced induced systemic resistance (ISR) in grapevine against the biotrophic oomycete Plasmopara viticola. By coupling flow cytometry with high-throughput 16S amplicon sequencing, this work provides a robust culturomics framework for screening stable microbial consortia and highlights scion-selected bacterial populations as effective drivers of induced grapevine resistance against P. viticola.",
