"abstract": "Background\n\nFilamin A (FLNA), a scaffolding protein extensively expressed in various tumor cells, is involved in regulating multiple cellular functions.Objective\n\nThis study aimed to investigate the role of FLNA in the occurrence, development, and prognosis of glioma.Materials and methods\n\nFirstly, data from public databases including The Cancer Genome Atlas (TCGA), Chinese Glioma Genome Atlas (CGGA), and Gene Expression Omnibus (GEO) were utilized to analyze FLNA expression in glioma, evaluate the impact of differential FLNA expression on the prognosis of glioma patients, and validate the findings using tissue samples from our institution. Subsequently, cell experiments were conducted to confirm the effect of FLNA on the malignant phenotype of glioma cells. Finally, differential gene expression analysis and Gene Set Enrichment Analysis (GSEA) were performed to explore the association between FLNA and downstream signal transduction pathways.Results\n\nIn the TCGA, CGGA, and GEO databases, FLNA expression was significantly higher in tumor tissues than in normal tissues, with a notably higher expression in glioblastoma multiforme (GBM) compared to lower-grade glioma (LGG). Survival analysis revealed a negative correlation between FLNA expression and patient survival. Receiver Operating Characteristic (ROC) curve analysis indicated that FLNA could serve as a prognostic indicator for glioma. Univariate and multivariate analyses identified FLNA as a high-risk factor influencing prognosis. In clinical samples from our institution, FLNA expression was significantly elevated in glioma tissues relative to adjacent non-tumor tissues. In vitro experiments demonstrated that glioma cells with high FLNA expression exhibited enhanced proliferation, migration, and invasion capabilities. Gene enrichment analysis suggested a potential association between FLNA and the Epithelial-Mesenchymal Transition (EMT) process.Conclusion\n\nFLNA is highly expressed in glioma and correlates with poor prognosis. It may enhance the proliferation, migration, and invasion of glioma cells through the EMT process, thereby emerging as a potential independent prognostic factor for glioma.",
