"abstract": "Background: Rheumatoid arthritis (RA) is a chronic inflammatory autoimmune disorder characterized by joint inflammation, pain, and progressive joint destruction. Despite advancements in conventional therapies, many patients with RA experience inadequate response or intolerance to synthetic drugs. Plant-based medicines have emerged as promising alternatives due to their diverse pharmacological properties and relatively fewer side effects. Among these, HXQF has been clinically used for treating various inflammatory conditions, including RA, with remarkable efficacy. However, the underlying mechanisms of its therapeutic actions remain elusive. Purpose: The primary objective of this study was to elucidate the pharmacological mechanisms of HXQF in the treatment of RA. By utilizing a multidisciplinary approach, we aimed to gain insights into its mode of action and provide a scientific basis for its clinical application. Study design: A comprehensive study design combining network pharmacology, molecular docking, molecular dynamics simulations, and pharmacological experiments was employed to investigate the mechanisms of HXQF. Methods: First, network pharmacology was used to identify potential targets of HXQF related to RA. Molecular docking was then performed to assess the binding affinity of HXQF components to these targets. Next, molecular dynamics simulations were conducted to explore the stability and dynamics of the protein-ligand interactions. Finally, pharmacological experiments were carried out to validate the predicted targets and mechanisms in vitro and in vivo. Results: Our findings revealed that HXQF exerts its therapeutic effects on RA through multiple targets and pathways. Notably, pharmacological validations suggested that certain components of HXQF, such as isoimperatorin (ISO), may play a significant role in reducing inflammation and mitigating joint damage. Conclusion: This work demonstrates that HXQF and its key component ISO can relieve short-term inflammatory and joint lesions in RA rat models. Bioinformatic simulation predicts that their anti-inflammatory performance may be correlated with EGFR-related JAK-STAT and PI3K-Akt pathways, which requires further protein and cellular experimental verification to confirm the regulatory mechanism.",
