"abstract": "Background: Reduction of thyroid hormone levels is known as hypothyroidism produces an alterable effect of bodily activities more willingly than a hormonal deficit. Aim of the Work: The present research was designed to compare the valuable influences of Nigella Sativa oil (NSO) and thyroxine on carbimazole-persuaded hypothyroidism. Materials and Methods: Forty rats were utilized and allocated into five groups. Group I (without treatment), included subgroups Ia and Ib were. Group II received distilled water, orally, included subgroups IIa and IIb. Group III was administered oral carbimazole, daily for three weeks (subgroup IIIa) and subgroup IIIb (recovered for an additional four weeks). All subgroups a and b were sacrificed after three and seven weeks, respectively. Group IV received carbimazole, like group III, followed by four weeks of daily oral NSO. Group V was given carbimazole like group III, followed by four weeks of oral thyroxine, daily. Serum levels of T3, T4, and TSH as well as oxidative markers in tissue; malondialdehyde and reduced glutathione were analyzed biochemically. In addition to histological examination, Caspase 3 and IL 6 immunohistochemical evaluations were performed. Results: Either NSO or thyroxine produced modifications effects in carbimazole-provoked histopathological changes of the thyroid gland, as demonstrated by a significant decrease in Caspase 3 immuno-expression, with more substantial regulating influence linked to thyroxine. In addition, both medications showed anti-inflammatory consequences (a decrease in immunohistochemical IL 6) and antioxidant capability (reduction in malondialdehyde and rise in reduced glutathione). Both NSO and thyroxine improved the thyroid function, nonetheless an additional impact related to thyroxine. Conclusion: Either NSO or thyroxine have an outstanding capability to minimize carbimazole- provoked negative consequences on the thyroid gland by regulation of oxidation-reduction state, as well as effective inhibition of inflammatory responses; thyroxine was more effective.",
