Abstract

Parahydrogen-induced polarization (PHIP) represents a cost-effective hyperpolarization technique providing dramatic increase of the NMR spectroscopy sensitivity. In particular, hyperpolarized metabolites are employed in various in vivo applications, including cancer diagnostics and grading. In this proof-of-concept study, we present an extension of the PHIP methodology to a key nitrogenous metabolite, the nitrate anion. We report an approach for the production of hyperpolarized nitrate solutions utilizing PHIP by means of the side-arm hydrogenation of propargyl nitrate ester. Through the sequential optimization of hydrogenation, polarization transfer, and hydrolysis procedures, we obtained hyperpolarized nitrate anion with 15 N polarization levels estimated as >2%.
