Abstract
Staphylococcus haemolyticus has emerged as a significant multidrug-resistant (MDR) nosocomial pathogen, yet the genomic diversity of its prophages - key drivers of bacterial evolution and horizontal gene transfer remains poorly characterized compared to S. aureus . In this study, we isolated and sequenced a novel temperate bacteriophage, IME1318_01, induced from the MDR clinical strain S. haemolyticus Sah1318. Transmission electron microscopy revealed morphology characteristic of the Siphoviridae family, featuring an isometric head and a non-contractile tail. Genomic analysis identified 61 open reading frames arranged in a functional modular architecture typical of staphylococcal prophages. To clarify its evolutionary position, we performed comprehensive phylogenomic analyses using Genome-BLAST Distance Phylogeny (GBDP) and proteomic tree reconstruction. These analyses supported the classification of IME1318_01 as a distinct species within the Azeredovirinae subfamily, forming a strongly supported monophyletic clade with Staphylococcus phage StB12. Biological assays indicated a latent period of approximately 20 min and a burst size of 56 PFU per cell, confirming efficient replication dynamics upon induction. These findings expand our understanding of the 'viral dark matter' in coagulase-negative staphylococci and offer valuable clues regarding the genetic mobility and host interactions of phages within antibiotic-resistant reservoirs.
