Abstract

Background: TiO2NPs toxicity has been studied extensively. Geraniol has shown protective potential against oxidative stress. Aim of the work was to evaluate toxicity of TiO2NPs on the brain of adult male albino rats and the potential protective effect of geraniol. Material and methods: 60 rats were allocated into 4 groups: Group I was subdivided into 3 equal subgroups; IA (-ve control group) (10 rats), IB (+ve control group; gum acacia) (10 rats): 1 ml of 5% gum acacia solution for each rat and IC (+ve control group; corn oil) (10 rats): 1ml corn oil for each rat, Group II (Geraniol group) (10 rats): 100 mg/kg geraniol dissolved in corn oil for each rat, Group III (TiO2NPs group) (10 rats): 1200 mg/kg TiO2NPs suspended in gum acacia solution for each rat and Group IV (TiO2NPs + geraniol group) (10 rats): each rat received geraniol followed 1 hour later by TiO2NPs at the same previous doses. All treatments were given orally once daily for 6 weeks after which all rats were sacrificed and brains were collected. Results: TiO2NPs caused significant decline of reduced GSH, dopamine and serotonin and significant increase of NO, 8 OH-dG, TNF-α and IL-1β with histopathological and immunohistochemical changes. Co-administration of geraniol caused a significant increase of reduced GSH, dopamine and serotonin and significant decrease of NO, 8 OH-dG, TNF-α and IL-1β with nearly complete regression of histopathological and immunohistochemical changes. In conclusion, TiO2NPs induced brain injury as demonstrated by biochemical and histopathological changes which were attenuated by co-administration of geraniol.
