Extracellular vesicle heterogeneity through the lens of multiomics

18 more authors

Abstract

Extracellular vesicles (EVs) are heterogeneous in size, biogenesis, content, and function.Aggressive cancer cells release a distinct, poorly characterized and particularly large EV subtype referred to as large oncosomes (LO). This study employs an optimized method to improve preparatory yields of LO and integrates mass spectrometry and RNAsequencing data to profile their molecular content. A consistent set of proteins enriched in LO is identified across glioma, prostate, and breast cancer cell lines. These species are also largely present as mRNA in LO from the prostate cancer model and abundant in plasma LO from patients with metastasis. Single LO RNA-sequencing confirms the findings from bulk LO cargo, demonstrating the utility of single-cell technologies for large vesicle analysis. These results demonstrate the feasibility of multiomic approaches to characterize EV composition, and further suggest the potential prognostic value of LO for developing a multi-analyte approach for liquid biopsy.
