Chemical Synthesis of the endogenous metabolite Phosphocitrate and Its Structural Isomer Phosphoisocitrate

Abstract

The first chemical synthesis of phosphoisocitrate [1-(phosphonooxy)propane-1,2,3-tricarboxylic acid, i-PC], a structural isomer of phosphocitrate (PC, natural regulator found in mammalian cells) is reported.Isocitrate is an essential metabolite within the cellular citric acid cycle.Although i-PC may form in vivo via the phosphorylation of isocitrate, it has remained uncharacterized in literature.Phosphocitrate [2-(phosphonooxy)propane-1,2,3-tricarboxylic acid, PC] synthesis was previously reported on a ~200 mg scale. Here an improved synthetic procedure for PC on a multi-gram scale is presented.
