Abstract

Pit and fissure sealants (PFS) are effective in preventing occlusal caries. This study aims to evaluate the shear bond strength (SBS), microleakage, and penetration depth of the newly developed bioactive PFS (Biocoat™) and a fluoride-releasing resin-based PFS (Fissurit FX) under in vitro conditions. A total of 120 sound permanent molars were included. Specimens were assigned to four surface preparation (SF) groups (Acid; Acid + Bond; Sodium hypochlorite (NaOCl) + Acid; NaOCl + Acid + Bond) and two PFS materials subgroups (BioCoat™; Fissurit FX). SBS was evaluated after artificial aging. Microleakage and penetration were evaluated using stereomicroscopic and dye penetration methods. The SF (p = 0.001) and aging (p = 0.018) had significant effects on SBS, whereas no difference was found between materials (p = 0.773). The highest SBS value was obtained with NaOCl + Acid SF (23.14 ± 7.73 MPa) and the lowest with NaOCl + Acid + Bond (12.50 ± 2.88 MPa). The SBS value before aging was 19.5 ± 6.45 MPa, while after aging it was found to be 15.16 ± 7.68 MPa. The SF and material type showed no significant effect on microleakage and penetration (p > 0.05). Pretreatment with 5.25% NaOCl before acid etching improved SBS, whereas no statistically significant differences were observed in microleakage or penetration. These findings suggest a potential mechanical advantage under laboratory conditions without indicating overall clinical superiority.
