Abstract
α-l-Rhamnosidase is a key biocatalyst for glycosidic bond hydrolysis. Icariside I is a high-value flavonoid glycoside with enhanced bioavailability. However, its production from icariin is limited by enzyme stability and suboptimal catalytic performance. Here, an α-l-rhamnosidase mutant (R783A) was heterologously expressed, purified, and immobilized onto UiO-66-NH2via a simple adsorption strategy to construct a robust biocatalyst. The resulting R783A@UiO-66-NH2 exhibited markedly improved stability, including broad pH tolerance (6.0–9.0), retained more than 90% of its initial activity after 2 h at 60 °C, enhanced storage stability (88.6% residual activity after 30 days), and improved resistance to metal ions and organic solvents. Notably, R783A@UiO-66-NH2 efficiently catalyzed the hydrolysis of icariin, achieving a 99.8% conversion and a 97.6% icariside I yield within 5 h, corresponding to a space-time yield of 0.52 g L−1 h−1. In addition, the immobilized biocatalyst retained 85.6% yield after five consecutive reuse cycles and 60.3% after eight cycles at 60 °C, demonstrating useful operational stability and recyclability. This study provides a simple and efficient approach for the biotransformation of icariside I, with UiO-66-NH2 nanoparticles offering a versatile platform for high-value flavonoid production.
