Abstract

By-products from shrimp processing are valuable sources of astaxanthin, a high-value carotenoid known for its bioactive properties. This study investigated choline chloride-based deep eutectic solvents (DESs) in combination with ultrasound-assisted extraction (UAE) to recover astaxanthin from banana shrimp (Fenneropenaeus merguiensis) by-products. Additionally, the study evaluated the antioxidant and ACE-inhibitory activities of the resulting extracts. Three choline chloride-based DESs, namely ChCl: urea, ChCl: glucose, and ChCl: ethylene glycol, were screened at a 1:2 M ratio with 10% water, among which ChCl: glucose showed the highest extraction efficiency, yielding 29.50 ± 0.04 μg/g astaxanthin. Ultrasound-assisted extraction markedly enhanced astaxanthin recovery, with the highest yield obtained after 15 min of ultrasound treatment at 200 W, reaching 50.10 ± 0.05 μg/g, approximately 70% higher than the control. However, higher ultrasound intensity or prolonged exposure did not further improve extraction, likely due to localized heating, oxidation, and carotenoid isomerization. Compared with the conventional acetone extract, the DES-derived astaxanthin-rich extracts exhibited competitive and concentration-stable bioactivities, with DPPH and ABTS radical-scavenging activities and ferric-reducing power ranging from 34.41% to 45.89%, 29.28% to 61.06%, and 0.109 to 0.148, respectively. ACE inhibitory activity ranged from 32.79% to 45.97%, with the highest inhibition observed after 10 min ultrasound treatment at 400 W. Overall, DES-UAE provided an efficient approach for recovering astaxanthin-rich extracts from banana shrimp by-products under the tested laboratory-scale conditions.
