Coupling Insoluble Pteroic Acid Using Quaternary Ammonium Salts: Synthesis of a Peptide Bearing a Folic Acid Moiety
- 1NIPPON SHOKUBAI CO., LTD, Nishi Otabi-cho, Suita, Osaka, 564-0034
Abstract
Herein, I report a unique coupling method for insoluble pteroic acid. Tetraalkylammonium hydroxide effectively enabled solubilization, efficient coupling with an amine, and an easy workup process to produce folate-conjugated peptides. The generality and versatility were confirmed through coupling studies of several amino acids, demonstrating its applicability to the synthesis of various folate and pteroate conjugates.
